Relevance to Autism

This gene has been associated with syndromic autism, where a subpopulation of individuals with a given syndrome develop autism. In particular, genetic association exists between autism and tuberous sclerosis (and hence the TSC1 and TSC2 genes as well).

Molecular Function

This gene encodes a growth inhibitory protein thought to play a role in the stabilization of tuberin and has been implicated as a tumor supressor.

Model Summary

Tsc1 mutant flies show impairments in olfactory associative learning as well as a severe decrease in reversal learning task, a measure of behavioral flexibility. This deficit seems to be due to enhanced old memories, driven by inability to active Rac1-dependent forgetting. This behavioral inflexibility seems to be driven by Tsc1 function in the mushroom bodies.
