Relevance to Autism

An inhibitor of topoisomerase (TOP1), topotecan, reduces the expression of extremely long genes in both mouse and human neurons in a dose-dependent manner; interestingly, the expression of many high-confidence ASD candidate genes is also reduced after TOP1 inhibition (King et al., 2013).

Molecular Function

This gene encodes a DNA topoisomerase, an enzyme that controls and alters the topologic states of DNA during transcription. This enzyme catalyzes the transient breaking and rejoining of a single strand of DNA which allows the strands to pass through one another, thus altering the topology of DNA.
